  • Systems pharmacology internship Modelling of Iron Metabolism

    Modelling of Iron Metabolism

    Project:

Iron is essential for life on Earth and critical for human health. It enables oxygen transport via haemoglobin, supports cellular energy production, and plays a role in enzyme function and detoxification. A lack of iron causes anaemia, leading to fatigue and weakness, while excess iron can result in organ damage.

Mechanistic mathematical modelling of iron metabolism could offer valuable insights for drug development, particularly in hematology. However, existing iron metabolism models found in the literature often lack technical comprehensibility and do not integrate most recent knowledge from mechanistic studies and publicly available datasets. The internship candidate will revisit existing iron metabolism models and integrate them with novel scientific insights using state-of-the-art modelling approaches.

    Internship goals:

    • Gaps or areas needing refinement based on current biological understanding and/ or technical considerations are identified.
    • A comprehensive structural model of iron metabolism (using ODEs) is developed.
    • Validated physiological parameters using Nonlinear Mixed Effects Modelling (with Monolix software) and in-house datasets compiled from literature are estimated.
    • Various physiological and pathological conditions, offering new perspectives for drug development in diseases like anaemia and cancer, are simulated.

    Requirements:

    • Bachelor’s or Master’s degree from ETHZ, EPFL or any other recognized university.
    • Rigorous, excellent analytical skills
    • Confident in understanding, using and numerically solving ordinary differential equations (ODEs)
    • Expert in scientific programming using R (or some other closely related scripting language)
    • Pro-active and independent work style
    • Proficient scientific communication skills
    • Proficient in oral and written English
    • Strong team player, good communication skills and ability to work with cross-cultural and diverse project teams and clients

    Languages: English (advanced).

  • Systems Pharmacology Internship Antibiotics Resistance

Simulation software for antibiotics effect prediction

    Project:

Antibiotics resistance is a huge issue that is also getting more public awareness due to its criticality. An important part of addressing this problem is the correct usage of antibiotics. For this one has to know if an antibiotic is effective against a certain bacterial strain and how it should be dosed. We are implementing a software platform that allows users to predict the probability of successful treatment based on the bacterial strain and the type of drug and the dose and regimen. This software should be accessible via a web interface that is programmed in R and Shiny.

    Internship Goals:

    1. Implement a mathematical model of antibiotics pharmacokinetics.
    2. Implement a mathematical model of antibiotics effect.
    3. Develop an interface in shiny for the mathematical models.
    4. Rigorous testing of the interface for usability.
